606nm Dichroic Mirror for Microscope is a high performance optical filter designed specifically for fluorescence microscopy applications. It precisely reflects light centered around 606nm, while efficiently transmitting wavelengths outside this band.
- It Reflects over 98% of light at 606nm, ensuring strong excitation signal separation. And it provide over 90% transmission above 620nm, insure a high emission signal transmission
- We use IAD hard coating for the dichroic filter. With multi dielectric layers, the filter is extremly durable and stable on the optical performance
- It is Optimized for 45° Angle of Incidence. Ensures precise spectral performance in typical microscope filter cubes or beam-splitting assemblies.
- For the 606nm dichroic mirror, we use fused silica as a substrate. Fused silica material is low autofluorescence. It is suitable to reduce background noise during sensitive fluorescence detection.

606nm Dichroic Mirror Microscope Applications
- Dynamic Tracking of mCherry and tdTomato in Live Cells
When red fluorescent proteins are expressed to label target proteins in live cells, 561nm laser is used for excitation and the emission occurs above 610nm. The 606nm dichroic mirror efficiently reflects the 561nm excitation light while transmitting fluorescence signals above 610nm. This significantly reduces excitation light leakage into the detector and improves the signal-to-noise ratio in live-cell imaging. - Immunofluorescence Detection with Cy3/Alexa Fluor 555
Cy3/Alexa Fluor 555 has an excitation peak between 550–565nm and an emission peak between 565–620nm. The 606nm dichroic mirror accurately separates the 561nm excitation light from the >610nm emission light, minimizing background interference. When used with a 605/70nm emission filter in widefield microscopy, it effectively suppresses tissue autofluorescence (common in the 500–600nm range) and enhances the contrast of weak signals. - In Vivo Imaging with Fluorescent Probes
Rhod-2 is excited by 552nm light and emits at 581nm, but >600nm signals are often collected to avoid cellular autofluorescence. The 606nm dichroic mirror efficiently transmits long-wavelength calcium signals, penetrates deeper tissue, and improves in vivo imaging signal-to-noise ratio, while precisely separating calcium signals from background noise. - FRET Imaging
In FRET pairs like CFP-YFP or GFP-mCherry, detection of acceptor mCherry excited by donor GFP is required. The 606nm dichroic mirror isolates the 488nm donor excitation light and only transmits mCherry’s FRET signal. Upon excitation with 488nm laser, the mirror reflects 488nm light to the sample and transmits signals above 606nm, purifying the mCherry signal.
